The space mission of John Jameson Jr. gives due to an invading alien individual. The rocket crashes in the middle of New York, causing Spider-Man to come to the rescue. Peter is filled with grief and sadness after the death of Gwen Stacy at the hands of the Hobgoblin, having nightmares and anxiety attacks. Meanwhile, with the help of the mysterious Chameleon, Harry Osborn, Gwen's killer escapes Ryker Island and hires mercenaries from various countries to capture Peter, including Kraven the Hunter, Silver Sable, Taskmaster and others to collect Peter's blood and cure him of a disease caused by the Hobgoblin serum, however, he can turn into something worse. Afraid that his job as Spider-Man will put someone in danger, Peter pushes people away, including his best friend Mary Jane and Aunt May, but he will discover that to survive the hunt, he will need to let people come in your life again. To make matters worse, an alien from Jameson's mission attaches itself to Peter, turning him into its host!
